Evaluarea genetica si monitorizarea factorilor moleculari si biotehnologici care influenteaza performantelor productive la speciile de sturioni de Dunare crescute in sisteme recirculante.

Obiectivul general al proiectului:

Identificarea, investigarea si monitorizarea factorilor biomoleculari si biotehnologici cu potential de influenta a performantelor productive a speciilor de sturioni de Dunare crescuti in acvacultura, in sisteme intensive recirculante, in vederea cresterii profitabilitatii si competitivitatii acestei activitati.

Rezumat romana:

Speciile de sturioni din Dunare sunt printre cele mai valoroase, deoarece pe piata globala exista o mare cerere atat pentru carnea, cat si pentru icrele lor. Din moment ce populatiile salbatice din Romania sunt la limita extinctiei, eforturile actuale se indreapta catre dezvoltarea unor sisteme de acvacultura, ca o masura alternativa pentru reducerea presiunii create de pescuitul excesiv. Obiectivul proiectului nostru este identificarea, investigarea si monitorizarea factorilor biomoleculari si biotehnologici cu potential de influenta a performantelor productive a speciilor de sturioni de Dunare crescuti in acvacultura, in sisteme intensive recirculante, in vederea cresterii profitabilitatii si competitivitatii acestei activitati. Atat genitorii utilizati pentru reproducere, cat si descendentii acestora obtinuti in primele generatii ca rezultat al incrucisarilor controlate in conditii artificiale vor fi caracterizati din punct de vedere fiziologic si genetic. Efectul diferitilor factori de stress (temperatura, hrana, etc.) din sistemele de acvacultura excesiva va fi evaluat la nivel fiziologic, biochimic si molecular si, ca si rezultat, vor fi evaluati parametri biotehnologici optimi necesari pentru imbunatatirea adaptabilitatii la stress, supravieturii si ratei de reproductivitate a sturionilor de Dunare in acvacultura. Liniile de descendenti avand calitati biologice superioare vor fi selectate pentru crestere si reproducere. Drept rezultat, va fi elaborata o tehnologie de crestere si reproducere a sturionilor de Dunare.

Rezumat engleza:

The Danube sturgeon species are among the most valuable species, since there is a large demand on the global market for both the meat and the caviar. Since the wild populations are almost depleted, the current efforts look towards developing the aquaculture system for sturgeons in Romania as an alternative measure to reduce the pressure through overfishing. Our project aims to identify, investigate and monitor the biomolecular and biotechnological factors with the potential to influence the productive performance of Danube sturgeon species, reared in aquaculture, in intensive recirculating systems, in order to increase the profitability and competitiveness of this activity. Danube genitors used for reproduction and their progeny obtained in the first generations as a result of the controlled cross-breeding under artificial condition will be characterized from genetic and physiological point of view. The effect of different stress factors (temperature, nutrients, etc.) in intensive aquaculture system will be evaluated at physiological, biochemical and molecular level and, as a result, the optimum biotechnological parameters required for the improvement of adaptability to stress, survival and rearing rates for the Danube sturgeons in aquaculture setting will be evaluated. The progeny lines with superior biological characteristics will be selected for feature exploitation. As a result, an improoved technology for reproduction and rearing of Danube sturgeons will be elaborated.

Rezultate estimate:

Etapa I: Optimizarea protocoalelor de lucru in vederea analizei markerilor moleculari cu scopul determinarii diversitatii genetice, identificarii speciilor si hibrizilor de sturioni si a markerilor biochimici si fiziologici corelati cu raspunsul la stres in conditii de acvacultura. Conceperea unui model tehnologic pentru cresterea, reproducerea si monitorizarea sturionilor in conditii artificiale.

Etapa II: Caracterizarea din punct de vedere genetic a genitorilor cu ajutorul ADNmt si microsatelitilor si selectia pe baza rezultatelor obtinute a loturilor de reproducatori pentru reproducerea controlata. Evaluarea indicilor de crestere si a performatelor biotehnologice in stadiile timpurii ale primelor generatii de descendenti obtinuti in urma reproducerii controlate.

Etapa III: Identificarea si analiza factorilor de stress (temperatura, metale grele, nutrienti, etc.) si a raspunsului indus de acestia prin metode biochimice si moleculare. Corelatia rezultatelor obtinute cu caracteristicile fenotipice si de crestere inregistrate si stabilirea parametrilor optimi de crestere si reproducere la sturionii din Dunare. Selectia materialului sturionicol cu grad superior de adaptabilitate in vederea explotarii ulterioare.

Etapa IV: Integrarea rezultatelor anterioare si selectia liniilor de reproducatori si descendenti cu caractersitici biotehnologice superioare. Elaboararea unei tehnologii de obtinere si crestere a sturionilor de Dunare cu caracteristici superioare.
